Purification and Properties of Bakers' Yeast Trehalase.

چکیده
Since recent surveys of insect hemolymph (1, 2) have shown that trehalose is the major blood sugar, the hydrolytic enzyme trehalase has also awakened the interest of investigators. A purified preparation of the enzyme from Galleria mellonella (3) and from Phormia regina (4) was recently obtained and some of its properties described. The biosynthesis of trehalose with an enzyme preparation from brewers’ yeast (5) and from bakers’ yeast (6) has been accomplished. One of us (7) has also determined the function of trehalose in bakers’ yeast as an energy source for cell division, compared to that of trehalose of insects, in which it serves as an energy source for flight. Yeast trehalase, however, has never been purified because the bulk of it remains strongly attached to the cell debris, and only a small part is soluble (8, 9). Autolysis, therefore, does not liberate enough enzyme to permit more than a 4to 5-fold purification. The aim of this report is to present a purification procedure for bakers’ yeast trehalase and to describe some of its properties.

Purification and Properties of the Catalase of Bakers’ Yeast
Catalase from bakers’ yeast has been purified to homogeneity in the analytical ultracentrifuge and in gel electrophoresis; sedimentation measurements permit an estimation of its molecular weight as 248,000. Under denaturing conditions, polyacrylamide gel electrophoresis revealed dissociation of a major component of molecular weight 61,000, which constituted 90% of the total protein of the stain...
متن کاملChanges in Trehalose Content of Baker's Yeast as Affected by Octanoic Acid
Octanoic acid inhibited ethanolic fermentation by Saccharomyces cerevisiae (bakers yeast) and the trehalose accumulation, however did not affect the endogenous degradation of trehalose. This inhibition may be explained by the binding of octanoic acid to hexokinase or other proteins of plasma membrane because they are not necessary for endogenous fermentation. The degradation of trehalose may be...
متن کاملFormaldehyde dehydrogenase from bakers' yeast.
In the course of studies of formaldehyde metabolism in yeast (I), an enzyme was found that catalyzed the oxidation of formaldehyde in the presence of glutathione and diphosphopyridine nucleotide. A similar enzyme was discovered independently by Strittmatter and Ball in liver (2). This paper describes the partial purification and some of the properties of yeast formaldehyde dehydrogenase. Some e...
متن کاملModulation of trehalase activity in Saccharomyces cerevisiae by an intrinsic protein.
The regulation of cytosolic trehalase activity in yeast has been described as cycles of activation by phosphorylation by cAMP protein kinase. In this paper, evidence is presented for another regulatory mechanism--the binding of an endogenous inhibitory protein. This negative modulator was isolated during the purification procedure of cytosolic cryptic trehalase from repressed wild-type cells of...
متن کاملEffect of Using Lactobacillus Brevis IBRC-M10790 and Baker’s Yeast on Phytate Degradation of Sourdough and Barbari Bread
The possible use of Lactobacillus brevis IBRC-M10790 with degrading phytase activity was investigated in sourdough and Barbari bread –using 20 and 30% of sourdough in the formulation- in order to enhance the nutritional bioavailability, and the efficiency of the bacteria in decreasing phytate via using in sourdough was compared to yeast. Results showed that L. brevis ...
